چکیده

delayed chondrocyte differentiation since Indian hedgehog (Ihh) is not expressed in the ulna of Ulnaless or Hoxa11//d11/ mice. Fibroblast growth factor 3 (Fgfr3), which is expressed at low levels in distal cells and at high levels in columnar chondrocytes, shows low expression in the Ulnaless and Hoxa11//d11/ zeugopod resembling the expression level in distal chondrocytes. However, the Unique cartilage matrix-associated protein (UCMA), a specific marker for distal chondrocytes, is only expressed in the outermost cell layers in ulna and radius of both mutant mouse lines. These results indicate that chondrocyte differentiation is arrested at an early step in Ulnaless and Hoxa11/d11 double mutant mice, before the differentiation of distal and columnar chondrocytes takes place.

Evidence that DmMANF is an invertebrate neurotrophic factor supporting dopaminergic neurons.

In vertebrates the development and function of the nervous system is regulated by neurotrophic factors (NTFs). Despite extensive searches no neurotrophic factors have been found in invertebrates. However, cell ablation studies in Drosophila suggest trophic interaction between neurons and glia. Here we report the invertebrate neurotrophic factor in Drosophila, DmMANF, homologous to mammalian MAN...

Downregulation of DmMANF in Glial Cells Results in Neurodegeneration and Affects Sleep and Lifespan in Drosophila melanogaster

In Drosophila melanogaster, mesencephalic astrocyte-derived neurotrophic factor (DmMANF) is an evolutionarily conserved ortholog of mammalian MANF and cerebral dopamine neurotrophic factor (CDNF), which have been shown to promote the survival of dopaminergic neurons in the brain. We observed especially high levels of DmMANF in the visual system of Drosophila, particularly in the first optic neu...

Characterization of the Structural and Functional Determinants of MANF/CDNF in Drosophila In Vivo Model

Mammalian MANF and CDNF proteins are evolutionarily conserved neurotrophic factors that can protect and repair mammalian dopaminergic neurons in vivo. In Drosophila, the sole MANF protein (DmManf) is needed for the maintenance of dopaminergic neurites and dopamine levels. Although both secreted and intracellular roles for MANF and CDNF have been demonstrated, very little is known about the mole...

Drosophila Neurotrophins Reveal a Common Mechanism for Nervous System Formation

Neurotrophic interactions occur in Drosophila, but to date, no neurotrophic factor had been found. Neurotrophins are the main vertebrate secreted signalling molecules that link nervous system structure and function: they regulate neuronal survival, targeting, synaptic plasticity, memory and cognition. We have identified a neurotrophic factor in flies, Drosophila Neurotrophin (DNT1), structurall...

Effects of new beta-type Ti-40Nb implant materials, brain-derived neurotrophic factor, acetylcholine and nicotine on human mesenchymal stem cells of osteoporotic and non osteoporotic donors

INTRODUCTION Treatment of osteoporotic fractures is still challenging and an urgent need exists for new materials, better adapted to osteoporotic bone by adjusted Young's modulus, appropriate surface modification and pharmaceuticals. MATERIALS AND METHODS Titanium-40-niobium alloys, mechanically ground or additionally etched and titanium-6-aluminium-4-vanadium were analyzed in combination wit...
